Newly Identified Motifs within PAS Domains of Filamentous Cyanobacteria

چکیده
Many cyanobacterial genome projects have been finished or are ongoing. Now, complete genome sequence of Anabaena sp. PCC 7120 (Anabaena)[1] and draft genome sequence of Nostoc punctiforme (Nostoc)[3] are available. Phylogenetically, these species are closely related. These species have extremely abundant signal-transduction proteins, especially PAS domain-containing proteins (PAScontaining proteins)[5]. PAS domain is known to be multi-functional domain superfamily with highly conserved 3D structure [6]. We formerly detected 142 and 180 PAS domains in 60 and 84 PAScontaining proteins from Anabaena and Nostoc, respectively. Clutering analysis showed that they were classified into 64 distinct subclasses and 36 “orphans”. On the other hand, we detected 35 pairs of PAS-containing proteins that are orthologous to each other between the two species. PAS domains in these orthologous poteins were classified into two groups: orthologous and non-orthologous ones. We interpreted the non-orthologous PAS domains in the orthologous proteins as generated by domain shuffling after branching of the two related species. Finally, 60 pairs of the orthologous PAS domains were highlighted as candidates for functionality [4]. Here, we extended our trial to unravel the complex PAS superfamily by finding novel motifs, which may interact each other in the 3-dimensional configuration.

p160/SRC/NCoA coactivators form complexes via specific interaction of their PAS-B domain with the CID/AD1 domain
Transcriptional activation involves the ordered recruitment of coactivators via direct interactions between distinct binding domains and recognition motifs. The p160/SRC/NCoA coactivator family comprises three members (NCoA-1, -2 and -3), which are organized in multiprotein coactivator complexes. We had identified the PAS-B domain of NCoA-1 as an LXXLL motif binding domain. Here we show that NC...
متن کاملGenomewide Analysis of Carotenoid Cleavage Dioxygenases in Unicellular and Filamentous Cyanobacteria
Carotenoid cleavage dioxygenases (CCDs) are a group of enzymes that catalyze the oxidative cleavage steps from carotenoids to various carotenoid cleavage products. Some ccd genes have been identified and encoded enzymes functionally characterized in many higher plants, but little in cyanobacteria. We performed a comparative analysis of ccd sequences and explored their distribution, classificati...
متن کاملPAS domains: internal sensors of oxygen, redox potential, and light.
PAS domains are newly recognized signaling domains that are widely distributed in proteins from members of the Archaea and Bacteria and from fungi, plants, insects, and vertebrates. They function as input modules in proteins that sense oxygen, redox potential, light, and some other stimuli. Specificity in sensing arises, in part, from different cofactors that may be associated with the PAS fold...
متن کاملPermanent draft genome of strain ESFC-1: ecological genomics of a newly discovered lineage of filamentous diazotrophic cyanobacteria
The nonheterocystous filamentous cyanobacterium, strain ESFC-1, is a recently described member of the order Oscillatoriales within the Cyanobacteria. ESFC-1 has been shown to be a major diazotroph in the intertidal microbial mat system at Elkhorn Slough, CA, USA. Based on phylogenetic analyses of the 16S RNA gene, ESFC-1 appears to belong to a unique, genus-level divergence; the draft genome se...
متن کاملCyanophycin Mediates the Accumulation and Storage of Fixed Carbon in Non-Heterocystous Filamentous Cyanobacteria from Coniform Mats
Thin, filamentous, non-heterocystous, benthic cyanobacteria (Subsection III) from some marine, lacustrine and thermal environments aggregate into macroscopic cones and conical stromatolites. We investigate the uptake and storage of inorganic carbon by cone-forming cyanobacteria from Yellowstone National Park using high-resolution stable isotope mapping of labeled carbon (H(13)CO3 (-)) and immun...
